Key backman Griffin Logue picked up a minor hamstring strain in Round 1 and won't feature against the Demons, while lockdown defender Jackson Archer will also miss after his three-match suspension for rough conduct was upheld on Tuesday night.
Dylan Stephens, Miller Bergman, Toby Pink, Finnbar Maley and Brynn Teakle have all been added to the Round 2 squad.
Charlie Comben faced a fitness test during Thursday's main training after sustaining a foot sprain against the Bulldogs. The 23-year-old remains in this side and is expected to face Melbourne.
Stephens, Bergman, Pink, Maley and Teakle are on the extended interchange bench along with Jacob Konstanty, Darcy Tucker and Will Phillips. The bench will be reduced to five players when the Roos announce their team at 5pm AEDT on Friday.
One interchange player will be named substitute when the final line-up is confirmed an hour before Sunday's 3.20pm bounce.
Recruit Jack Darling will celebrate his 300th AFL game against the Demons, becoming the 106th player in VFL/AFL history to achieve the feat.
